Figure 2.Confocal microscope analysis of internalization mAb J591.LNCaP cells were incubated with mAb J591 and FITC-transferrin (A,C,E) or mAb 7E11-C5.3 and FITC-transferrin (B,D,F) for 2 hrs and processed for immunofluorescence as described in Materials and Methods. mAb J591 (A) and 7E11-C5.3 (B) detected with Texas-Red conjugated secondary antibody. FITC-transferrin uptake (C and D). Images in A and C are merged to obtain image E (mAb J591 and FITC-transferrin co-localization, yellow). Images in B and D are merged to obtain image F where only transferrin uptake is seen since 7E11-C5.3 neither binds nor internalizes.
